Using the Spectronic 20 Spectrophotometer

  1. Turn instrument on (using the dial on the left) and wait 15 minutes to allow it to warm up.
  2. Set the desired wavelength by adjusting the dial on the top of the Spec 20.
  3. Adjust the left dial until the transmittance reads 0%.
  4. Wipe the outside of a "blank" cuvette containing distilled water and insert the cuvette into the cell compartment. Make sure that the etching on the cuvette lines up with the line on the compartment.
  5. Adjust the right dial until the transmittance reads 100%.
  6. Remove the blank and insert a cuvette containing the sample to be analyzed. Make sure to tap out any bubbles and wipe the outside of the cuvette before placing in the Spec 20. Also, line up the etching on the cuvette with the line on the compartment.
  7. Read the % transmittance and absorbance.
  8. If testing more than one sample, make sure to reset the Spec 20 using the blank in between each reading.
